Description

PCBIS (www.pcbis.fr) aims at giving access to assay development, screening facilities, early ADMETox services and to a collection of small molecules in order to develop either research tools or starting points for drug candidates. We got ISO 9001 certification (2007) and the French label of certification NF X50-900 (2014). Initiated in 1999 PCBIS is now a CNRS/University of Strasbourg labelled laboratory “UAR 3286”. We are IBiSA labelled at the national level, and part of the "Institut du médicament de Strasbourg" and of the national research infrastructure "ChemBioFrance". Our lab is divided in several functional units:

  1. Chemical Library Unit: it is in charge of the collections of small molecules (French chemical library: >80 000 compounds; Prestwick chemical library: 1520 drugs; Strasbourg chemical library: 8000 compounds; fluorescent chemical libraries: > 3000 compounds). This includes management, annotations and quality control of libraries. With 2 engineers (Dr B. DIDIER (CNRS) and Dr C. MARSOL (Unistra)) we can ensure a professional monitoring and quality control of libraries, realize a structure relationship analysis activity and participate to the synthesis of compounds after screening campaigns, thanks to the close collaboration with medicinal chemists (Didier Rognan’s lab). At this level PCBIS specificity consists in having direct access to competencies in medicinal chemistry, and having access to competencies in unique fluorescent chemical libraries and fluorescent probes which allow us to set up original assays for researchers.
  2. Assay and Target Production Unit: it is in charge of development, production & validation of molecular targets & cellular models before their use for screening. We also develop & validate assays. With 3 engineers (C. VALENCIA, V. Calco (CNRS) & R. HANY (fixed-term contract)) we develop models with researchers aiming to use chemical biology strategy. PCBIS can also develop unique assays a) assays for orphan membrane receptors (using fluorescent libraries), b) assays independent of the function for soluble proteins using fluorescent libraries, or (when available) using known ligand modified to become fluorescent, c) specific assays using other technologies. We also develop organoids in order to decrease the number of animals used for preclinical assays.
  3. High Throughput Screening Unit: it takes care of miniaturization, automation and validation of assays. With an engineer (S. GIORIA (CNRS)) and 2 technicians (A. OBRECHT & C. LEHALLE (Unistra)) we miniaturize and validate screening assays, perform screening campaigns of chemical libraries on molecular and cellular assays in order to provide validated hits. We are able to propose a large panel of technologies using different luminescent & fluorescent (FRET, anisotropy, HTRF,…) or label free techniques (EPIC) in binding and functional assays.
  4. ADME-Tox Unit: it evaluates the early ADMETox properties of active molecules. This consists to determine absorption, distribution, metabolism, elimination & toxicity of molecules. It is necessary to determine or to predict these properties as early as possible to avoid failures and to limit costs in chemical synthesis, in vivo studies and formulation. With 4 engineers (Dr P. GIZZI (CNRS), Dr F. DAUBEUF (Unistra), C. BOURBAN, C. WAGNER) we are able to propose a large panel of services in this field, including physico-chemical properties measurements and in vitro and in vivo evaluations of compounds.
  5. Microfluidics unit: development of microfluidic tools for research and organ on a chip. With an Engineer (C BRAINI)
  6. Training activities: supervise students or researchers; to participate in training or courses in drug discovery strategies, HTS, ADMETox. We also participate to professional training session for students and people coming from industry (3-4 sessions per year).
